Cytology preparation

  • How are cytological specimens prepared?

A drop of sample is placed near the frosted end of a glass slide (a).
A second slide is inverted over the sample, making gentle contact with the sample (b).
The slides are pulled apart, maintaining even contact (c), resulting in 2 evenly covered slides with cellular material concentrated in the center (d)..

  • How do you prepare a cell block in cytology?

To prepare the cell block, the sample is aliquoted into the collodion lined tube and then centrifuged for 6-10 minutes at 1600-2500 rotations per minute (RCF 1125 g).
The supernatant is then carefully decanted or pipetted off (or can be removed later as the bag is being cut)..

  • What are the procedures for cytology?

Some of the ways a provider can collect cytology test samples include:

Brushing or scraping tissue from the surface of a part of your body.Collecting fluid or discharge samples from your body, such as a pee sample.Using fine-needle aspiration to draw a fluid sample from an area in your body..

  • What is a cytology procedure?

Cytology is the exam of a single cell type, as often found in fluid specimens.
It's mainly used to diagnose or screen for cancer.
It's also used to screen for fetal abnormalities, for pap smears, to diagnose infectious organisms, and in other screening and diagnostic areas..

  • What is cytological preparation?

In the cytology laboratory various sample preparation steps are performed prior to analysis including centrifugation to concentrate cells in dilute samples, followed by preparation and staining of slides, and examination under a light microscope (Figure 1)..

Use a saline moistened swab (sterile saline, not water, as the osmotic effect of water ruptures the cells) to remove superficial debris. If bloody, swab/ dab with clean paper towel to remove blood first. Gently touch the tissue surface to a clean slide. Repeat touch imprint along the length of the slide.
In general, aspirate smears, needle rinses and cell blocks, alone or in combination, are prepared from a given sample.

How do you choose a cytopreparation method?

The method of cytopreparation may be selected on the basis of the physical properties of the specimen and the patient’s clinical history.
For example, highly viscous specimens may require a smear technique or mucolytic agents, and bloody specimens will benefit from methods that segregate larger epithelial cells from red blood cells.

What types of preparations are available for serous fluid cytology?

There are a variety of preparation types that are available for preparation of serous fluid cytology.
These include:

  1. direct smears and cytocentrifugation spins
  2. either Papanicolaou and/or modified Giemsa stained or ThinPrep® and SurePath™

Cell block preparations can be made from serous effusions.
