I - Structure and Catalysis The tertiary structure of a single chain protein is the overall 3D structure of the protein. A protein of a given primary structure folds to form a 3D structure with embedded secondary structures, super secondary structures and domains.
Tertiary structure is the overall 3D structure of the protein. Quaternary structure is the overall structure that arises when separate protein chains aggregate with self to form homodimers, homotrimers, or homopolymers OR aggregate with different proteins to form heteropolymers.
Secondary structure is determined by Hydrogen bonds between the backbone of the chain. Tertiary structure is determined by all electrostatic interactions (e.g. H-bond, Van der Waals) as well as disulfide bridges. Quaternary structure is determined by the subunits and the attractions between the different subunits.
However, some proteins are made up of multiple polypeptide chains, also known as subunits. When these subunits come together, they give the protein its quaternary structure. We’ve already encountered one example of a protein with quaternary structure: hemoglobin.
