oct freezing medium

Is it OK to freeze tissues in Oct media?

These tissues were cryopreserved in 30% sucrose before being placed in OCT and frozen in dry ice/100% ethanol. If freeze/thaw will cause too much damage, I could just mount the tissues in what I think is the right position and section. Why we soak tissues in 30% sucrose prior to embedding in OCT media?

What is tissue freezing medium?

Tissue Freezing Medium; TFM™. An embedding matrix for frozen sections. TFM™ is an ultra pure formulation of water-soluble glycols and resins that provides a solid bond between the tissue and the object holder with the following features: Less Ice Artifacts: TFM's reduced water content minimizes freeze-fracturing.

Can you freeze Oct cells if submerged in liquid nitrogen?

Note: DO NOT freeze the tissue by submersing into the liquid nitrogen, it will cause the OCT blocks to crack which makes them very difficult or impossible to section. This happens because the outside tissue begins to freeze much more quickly than internal portion.

What is the most common freezing medium?

The most common freezing medium is 90% fbs/10% dmso. 1 thaw fbs, dmso, prepare freezing medium, 70% dmem, 20% fbs, 10%. The composition of the different freezing media tested is shown in table 1. Materials · freeze medium or glycerol ) · 70% alcohol in sterile water · pbs without ca2+/mg2+ .
