  • Why is glutaraldehyde used for cell fixation?

    Besides its application as disinfectant and medication, glutaraldehyde is used in biomedical research to fix cells.
    The principle behind the fixation is the binding of glutaraldehyde to nucleophiles of which the amino groups are the most abundant but binding to, e.g., sulfhydryl groups also occurs (Griffiths, 1993).

  • Proteins.
    Glutaraldehyde cross-links proteins in a manner analogous to formaldehyde, but targeting different components, such as amino groups.
    During this process, it stabilizes the protein mass and preserves the morphology.
    It fixes somewhat faster than formaldehyde, but its penetration rate is poorer.

  • How do you prepare glutaraldehyde fixation solution?

    Glutaraldehyde fixation protocol.
    Solution Recipes: 1M phosphate buffer made as follows: Add 68.4 mLs of 1M Na2HPO4 + 31.6 mLs of NaH2PO4 to 900 mLs dH20, pH should be right around 7.2, doesn't hurt to check.

  • Fixation of Tissue
    1. For immersion fixation, use 2.5% glutaraldehyde (must be EM grade) in 0.1M buffer.
    2. For perfusion fixation, use 2% glutaraldehyde and 2% paraformaldehyde in 0.1M buffer.
    3. To prepare 100 mL of glutaraldehyde/paraformaldehyde:
