Why do we use 10% formalin for fixation?
The fixative 10% buffered formalin is commonly used to preserve tissues for routine histology in many labs.
The formaldehyde has a greater chance for oxidation in this concentration of tissue fixative and eventually the solution will start to drop in pH, in spite of the buffer.Fixation Protocols

What is the process of formalin fixation?
Formaldehyde fixes tissue by cross-linking the proteins, primarily the residues of the basic amino acid lysine.
Its effects are reversible by excess water and it avoids formalin pigmentation.
Paraformaldehyde is also commonly used and will depolymerize back to formalin when heated, also making it an effective fixative.
- If cells are grown on culture plates, grow to 80% confluence (50 million cells).
