  • Why do we use 10% formalin for fixation?

    The fixative 10% buffered formalin is commonly used to preserve tissues for routine histology in many labs.
    The formaldehyde has a greater chance for oxidation in this concentration of tissue fixative and eventually the solution will start to drop in pH, in spite of the buffer.

    1. If cells are grown on culture plates, grow to 80% confluence (50 million cells).
    2. Centrifuge the cells at 1,000-3,000 RPM for 1 minute.
    3. Resuspend the cells in 20 ml PBS and repeat step #2.
    4. Resuspend the cells in 20 mls 10% Neutral Buffered Formalin (NBF)(VWR, Cat.
