  • How long can tissue be fixed in formalin?

    All tissue specimens should be fixed in 10% neutral buffered formalin for a minimum of 3 days (72 hours) and up to 2 weeks (14 days) for biopsy or autopsy tissue, or up to 4 weeks (28 days) for brain autopsy tissue.

  • What is the duration of formalin fixation?

    Time of fixation: 6-18 hrs for biopsy specimens and 24-72 hrs for standard samples.
    Both under and over fixation alter the quality of molecular analytes.
    A recent report recommends that tissue not be fixed for more than 36hrs to avoid overfixation and excessive cross-linking.

  • Is formalin fixation time greater than 6 hours?

    The current guidelines of the American Society of Clinical Oncology and the College of American Pathologists (ASCO/CAP) on immunohistochemistry for breast cancer biomarkers recommends that specimens for histology should be fixed in 10% neutral buffered formalin for a minimum of 6 hours and a maximum of 72 hours.27 jan. 2021

  • Specimens should be fixed for approximately 6 to 72 hours,5 preferably for a minimum of 8 hours especially for larger specimens. “Overnight” fixation (i.e. 8-12 hours) is generally indicated for 10 mm thick slices of tissues.
    Fixation for 12-24 hours is considered optimal for most immunohistochemistry.

The most recent ASCO/CAP Guidelines related to HER2 testing in breast cancer recommendations, cytologic, biopsy and resection specimens should be placed in formalin in a timely fashion (within 1 hour) and should be fixed in 10% neutral buffered formalin for 6 to 72 hours.
