What is the cell fixation protocol with methanol?
Cells are usually plated one day prior to staining in order to achieve 60-80% confluency.
Fix the cells with 100% methanol for 10 minutes at -20°C.
Note: optimal fixation time and reagent depends on the antigen of interest and must be optimized.How long does it take to fix cells in methanol?
In a standard fixation protocol, ice-cold methanol is added to cells for 10-20 minutes at 4˚C.

How does methanol work as a fixative?
The fixation obtained with methanol is very similar to that of ethanol.
It is a non-additive precipitant fixative.
It fixes proteins by dehydration and precipitation.
Mixtures incorporating it are usually water free.
- Carefully cover cells with methanol fix and incubate for 5 min at -20°C.
- Wash three times with PBS for 10 min.
- Incubate with blocking buffer for 30 min.
- Incubate in incubation buffer containing the primary antibody (for appropriate dilution, refer to the data sheet) for 2 h at RT.
