  • The fixative 10% buffered formalin is commonly used to preserve tissues for routine histology in many labs.
    The formaldehyde has a greater chance for oxidation in this concentration of tissue fixative and eventually the solution will start to drop in pH, in spite of the buffer.

  • What is the duration of formalin fixation?

    Time of fixation: 6-18 hrs for biopsy specimens and 24-72 hrs for standard samples.

  • How do you prepare a 10% solution of formalin as fixative?

    Where only a standard stock solution of formalin* is available it is typically 37-40% formaldehyde (a gas) in aqueous solution and unbuffered.
    To make a histological fixative from this we need a 10% solution** of this stock formalin i.e. 1 part of the stock formalin with 9 parts water, preferably distilled.

  • How long to fix tissue in 10% formalin?

    All tissue specimens should be fixed in 10% neutral buffered formalin for a minimum of 3 days (72 hours) and up to 2 weeks (14 days) for biopsy or autopsy tissue, or up to 4 weeks (28 days) for brain autopsy tissue.

  • -We generally fix in 10% neutral buffered formalin (NBF) for at least 48hrs. It is good for basic histology, most special stains, and can also be used for many IHC protocols.
