1 to 1 serial dilution
What is a ten-fold serial dilution?
Figure 1. A ten-fold serial dilution, which can also be called a 1:10 dilution, or a series with dilution factor of 10. To determine the concentration at each step of the series, you divide the previous concentration by the dilution factor. *Dilution tubes begin with 9-mL. 1-mL is added and mixed, then 1-mL is transferred to the next tube.
What determines the size and number of dilution steps required?
The initial concentration and target range needed determines the size and number of dilution steps required. Serial dilutions are often performed in steps of 10 or 100. They are described as ratios of the initial and final concentrations. For example, a 1:10 dilution is a mixture of one part of a solution and nine parts fresh solvent.
How do you calculate serial dilution?
In serial dilutions, you multiply the dilution factors for each step. The dilution factor or the dilution is the initial volume divided by the final volume. DF = V_i/V_f For example, if you add a 1 mL sample to 9 mL of diluent to get 10 mL of solution, DF = V_i/V_f = (1"mL")/ (10"mL") = 1/10. This is a 1:10 dilution.
What is the final dilution factor?
The final dilution factor of the fourth tube in your serial dilution is 1:10,000. The concentration of your substance is now 10,000 times less than the original undiluted solution. Determine the concentration of the solution following dilution.
Serial Dilution Objectives
The objective of the serial dilution method is to estimate the concentration (number of organisms, bacteria, viruses, or colonies) of an unknown sample by the enumeration of the number of colonies
Serial Dilution Formula and Calculations
Serial dilution involves the process of taking a sample and diluting it through a series of standard volumes of sterile diluent, which can either be distilled water or 0.9 % saline. Then, a small measured volume of each dilution is used to make a series of pour or spread plates.Depending on the estimated concentration of cells/organisms in a sample, the extent of dilution is determined. For e.g., if a water sample is taken from an extremely polluted environment, the dilut

Procedure of Serial Dilution
The following is the procedure for a ten-fold dilution of a sample to a dilution factor of 10-6: 1. The sample/culture is taken in a test tube and six test tubes, each with 9 ml of sterile diluents, which can either be distilled water or 0.9% saline, are taken. 2. A sterile pipette is taken. 3. 1 ml of properly mixed sample/culture is drawn into th
Applications of Serial Dilution
Serial dilution is performed in a number of experimental sciences like biochemistry, pharmacology, physics, and homeopathy. 1. Serial dilution is used in microbiology to estimate the concentration or number of cells/organisms in a sample to obtain an incubated plate with an easily countable number of colonies. 2. In biochemistry, serial dilution is
Limitations of Serial Dilution
Even though serial dilution is a useful technique in laboratories, it faces some challenges. Some of which are: 1. An error might occur during the propagation of the sample, and the transfer inaccuracies lead to less accurate and less precise transfer. This results in the highest dilution to have the most inaccuracies and the least accuracy. 2. Bec
