  • Why is iodine used to detect starch hydrolysis?

    The iodine is used as an indicator to show whether or not the starch has been hydrolyzed.
    Iodine combines with the intact starch to produce a blue-black colored complex.
    When then starch is hydrolyzed by the amylase, the simple sugars do not complex with the iodine and the medium remains clear and colorless.

  • Is hydrolyzed starch positive in iodine test?

    Here the starch is completely hydrolyzed which tells us that the starch is completely converted into Mono-saccharides, Di-saccharides, or Tri-saccharides.
    These saccharides give a negative Iodine test.

  • What is the acid hydrolysis test for starch?

    In acid hydrolysis methods, the starch is hydrolysed to glucose which is subsequently assayed.
    Dried homogenous food samples are dispersed in hot Ethanol solutions.
    The monosaccharides and oligosaccharides in the sample are soluble in the ethanol solution, while the starch is insoluble.

  • Areas where starch has been digested by bacterial growth exhibit clear halos in the midst of the dark plate, indicating a positive alpha-amylase, or starch hydrolysis test.
    Plates containing bacteria without alpha-amylase are uniformly dark, a negative result.

In order to interpret the results of the starch hydrolysis test, iodine must be added to the agar. The iodine reacts with the starch to form a dark brown color. Thus, hydrolysis of the starch will create a clear zone around the bacterial growth.
