cryosectioning troubleshooting
How do you fix cryosections?
Fix sections in cold acetone (-20°C) for 2 min.
Dry fixed slides completely (usually 1 hour at room temperature).
Store in a -70°C freezer until use. What is the best temperature for a cryostat?
General Approaches in Surgical Pathology
A cryostat is a microtome machine for cutting tissue at low temperatures (typically around − 15 to − 30 °C) (Figure 55).How long does it take for a cryosection to dry?
Note: The suggested cryostat temperature is between -15 and -23 °C.
The section will curl if the specimen is too cold.
If it is too warm, it will stick to the knife.
Air dry the frozen sections for 30 minutes at room temperature to prevent sections from falling off the slides during antibody incubations.- "Tissue doesn't stick to slide"--the temperature difference helps the tissue melt and stick to the glass.
Don't leave the glass slide in the crystat.
Breathe on the slide or touch it on the back to increase its temperature before lowering it into the cryostat.
