What is DIG-labeled RNA used for?
DIG-labeled RNA is used in a variety of hybridization techniques: The DIG RNA Labeling Mix is especially designed for the use with SP6,T7 and T3 RNA polymerases from Roche which are supplied with an optimized transcription buffer. Function tested in the DIG RNA Labeling Kit and in the DIG Nucleic Acid Detection Kit. For life science research only.
How is dig incorporated into RNA?
DIG is incorporated into the RNA at approximately every 25-30 nucleotides. The DNA template to be transcribed is cloned into the polylinker site of an appropriate transcription vector, which contains promoters for SP6 and or T3 and T7 RNA polymerases.
How is DIG-labeled RNA synthesized?
In the standard reaction about 10 g „full length“ DIG-labeled RNA is synthesized from 1 g linearized plasmid DNA with an insert of approx. 1 kb in 2 h. Larger amounts of DIG-labeled RNA can be obtained by scaling up the reaction components.
