L'interprétation repose sur la distinction de l'isotype : la présence d'IgM évoque une infection à un stade précoce ; les IgM sont encore présentes en début de phase secondaire mais absentes en phase tertiaire ; les IgG apparaissent en fin de phase primaire et sont présentes à titre élevé en phase tertiaire.
