Cytology sample collection and preparation

  • How do you prepare smear of cytological samples?

In general, diagnostic cytology is based upon three basic sampling techniques : (1) the collection of exfoliated cells, (2) the collection of cells removed by brushing or similar abrasive techniques, (3) the aspiration biopsy or removal of cells from non-surface –bearing tissue (or deep organs) by means of a needle, .

  • What are the preparation methods for cytology?

It is recommended that the aspiration is done with a syringe with air (2mL).
Smears are prepared from a small drop of the semisolid aspirate placed on a glass slide.
Advance the plunger of the syringe to express a small drop of the aspirated material on the center of the glass side..

  • What are the sampling techniques for diagnostic cytology?

We offer cytologic evaluation of multiple fluid types, including body cavity fluids, cerebrospinal fluid, synovial fluid and tracheal and bronchoalveolar washes.
Fluids from cystic masses and other sites (urine, bile) may also be submitted..

  • What is cytology collection?

Cytology: A specimen is ordered as a "cytology" when no core biopsy has been obtained.
This is when the aspirate is used to make cytology smears and placed in the cytojar ONLY.
Examples of cytology specimens include fine needle aspirations of thyroid, pancreas, cysts etc..

  • Scrape or brush cytology
Another way to get cytology samples is to gently scrape or brush some cells from the organ or tissue being tested. (This is also sometimes called a brush biopsy.) An example of a cytology test that samples cells this way is the Pap test.
Cytology sample types and collection
  1. Fine Needle Aspirates (FNA) This technique is non-invasive, generally non-painful, quick, and simple.
  2. Impression smears.
  3. Scrapings.
  4. Cytobrush Smears.
  5. Body Fluids.
  6. CSF Collection.
  7. Pleural, Peritoneal, Pericardial Fluid Sample Handling.
  8. Synovial Fluid Collection.
Sample collection and smear preparation Cytology samples can be collected from solid lesions by several techniques including: Aspiration cytology – fine needle aspiration (FNA): Ideal for cutaneous or subcutaneous masses since it avoids surface contamination.

How do I submit a cytolyt specimen?

*An optimal specimen would include:

  1. both submitting a specimen in tube and on slide

Aspirate specimen and place directly into a vial, tube or other container that contains ThinPrep® CytoLyt Solution or Saline. (This fluid is available by calling the Mercy Clinical Laboratory to arrange for delivery by courier).
